YPD Complex media for general growth and freezing
For 1 liter start with 900ml then add
10g Yeast Extract
20g Peptone
Autoclave
Add 100ml 10x Dextrose (final 2% dextrose = glucose)
Can add Zeocin if needed

BMGY Buffered complex media to generating biomass and for freezing
For 1 liter start with 700ml then add
10g Yeast Extract
20g Peptone
Autoclave
Add 100ml 1M Potassium Buffer pH 6.0
Add 100ml 10x YNB
Add 2ml 500x Biotin
Add 100ml 10x Glycerol (10xGY is 10% Glycerol so final 1% Gy)

BMMY Buffered complex media for expression of secreted protein
For 1 liter start with 700ml then add
10g Yeast Extract
20g Peptone
Autoclave
Add 100ml 1M Potassium Buffer pH 6.0
Add 100ml 10x YNB
Add 2ml 500x Biotin
Add Methanol to 0.5% final concentration

10x YNB10x Yeast Nitrogen Base
134g Yeast Nitrogen Base (with ammonium sulfate without amino acids)
Add water to 1 Liter and filter sterilize
Store 4°C

500xB 0.02% Biotin
Dissolve 20mg biotin in 100ml water and filter sterilize
