Protocol
- Centrifuge 1.5ml of overnight culture 20 seconds at full speed
- Pour off LB, Vortex to resuspend pellet
- Add 300µl TENS buffer, vortex 2-3 seconds
- Add 150µl 3M Sodium Acetate pH 5.2, vortex 2-3 seconds
- Centrifuge 2 minutes at full speed
- Pour supernatant into fresh tube
- Add 900µl Ethanol
- Centrifuge 2 minutes at full speed
- Wash pellet with 70% Ethanol then air-dry
- Resuspend in TE+RNAse
TENS
- 10mM Tris pH 8.0
- 1mM EDTA
- 100mM NaOH
- 0.5% SDS
