Degenerate Primer PCR

• Try to be not too degenerate, ≤ 65000 possibilities.
  
• Use Inosine to represent N, where all 4 residues are present
  EXCEPT for last 4 residues on 3’ end of primer
  
• Increase primer concentration by 10 to 20 fold
  
• Increase annealing time from 0:15 to ~0:45 or more
  
• Make control reactions with only 1 primer
