Antibiotics in Bacterial Plates

Ampicillin: Black stripe down side of plates

-After autoclaving 500ml LB/agar, cool to ~60'C in water bath

-Add 1 0.5ml Aliquots of 200mg/ml Ampicillin Stock

-Mix well by swirling bottle - avoid bubbles

-Pour plates

Tetracycline: Orange stripe down side of plates

-After autoclaving 500ml LB/agar, cool to ~60'C in water bath

-Add 1 500ul aliquot of 15mg/ml Tetracycline Stock

-Mix by swirling bottle - avoid bubbles

-Pour plates

-Store in Dark, wrapped in Aluminum Foil

-Does not work in presence of Magnesium

Chloramphenicol: Blue stripe down side of plates

-After autoclaving 500ml LB/Agar, cool to ~60'C in water bath

-Add 1 500ul aliquot of 34mg/ml Chloramphenicol Stock

-Mix by swirling bottle - avoid bubbles

-Pour plates

-Carcinogenic, avoid all contact with skin

Kanamycin: Purple stripe down side of plates

-After autoclaving 500ml LB/agar, cool to ~60'C in water bath

-Add 1 500ul aliquot of 10 mg/ml Kanamycin Stock

-Mix by swirling bottle - avoid bubbles

-Pour plates
