Precipitating DNA

Sodium Acetate Method

-Use most of the time.

-Use 3M Sodium Acetate at pH 4.5 for sequencing,

or pH 5.6 for most other uses.

-Add 1/10 volume 3M Sodium Acetate,

final concentration is 0.3M

-Add 2.5 volumes 100% Ethanol, allow to precipitate at RT

-Centrifuge

Ammonium Acetate Method

-Use when less protein desired

-Use 5M Ammonium Acetate

-Add 0.4 volumes 5M ammonium acetate

2M final concentration

-Add 2 new volumes Isopropanol, allow to precipitate at RT

-Centrifuge

 

Normally, 10-15 minutes centrifugation is fine.

If DNA is at less than 1ug/ml, then allow overnight precipitation and

increased centrifugation times.
